At present, most of the bags used for cultivating Pleurotus eryngii rely on traditional inoculation techniques, such as single-point and double-point inoculation. However, these methods—particularly manual bagging—often result in uneven mycelial growth both inside and outside the bags. To address this issue, it is essential to refine the inoculation process for better consistency and efficiency.
One effective approach is the seedling inoculation method. In this technique, the fungal strain is carefully inserted into the culture material at a depth of 2 to 3 centimeters. The hole is completely filled without leaving any gaps, ensuring optimal contact between the mycelium and the substrate. Each bag is inoculated at four different points, and then sealed with a ventilated plug. This method allows the mycelium to spread evenly from both ends of the bag, creating a full, uniform growth pattern throughout the entire bag.
Studies have shown that using the seedling inoculation method results in faster mycelial colonization compared to conventional techniques. For instance, the same type of bacterial bag can be fully colonized up to seven days earlier when using this method. Additionally, it is easier to handle and reduces the risk of contamination. As a result, Pleurotus eryngii can be inoculated either once or twice, depending on the desired outcome.
It is crucial to ensure that the bags are tightly sealed when loading the culture materials. Whether the inoculation is performed inside an inoculation box or in a dedicated area, all tools and surfaces must be properly disinfected and sterilized before use. After the bacteria are introduced, the bags should be sealed immediately to maintain a clean environment and prevent unwanted microbial contamination. These precautions help ensure successful cultivation and high-quality mushroom production.
